I looked at the Dio's user's list not long ago. In the last month, approx. 205 separate accounts logged into the site. In the past two months, the number was around 260. I honestly doubt there are many "regular players" who don't read Dio's at least once every two months. So, personally, I'd estimate that there are no more than 260 regular players tops. For every regular player who doesn't read dio's, there are people who read dio's who aren't regular players.
If you look at how many hours are logged on CF per day, then divide by the avg. number of hours played per day per player, that should give a good number. Going by the current users graph (at the time of this posting), the total hours for the trailing 24 hours is 815. That's just adding the max players per hour, for all hours in the period. Unfortunately, what we want is the *average* number of players logged in during each one hour period, which is going to be somewhat less than the max. If we guess that it's about 5 less, on average, then that puts the hour count at about 700. The question is how many hours per day the average CF player logs. I'd say somewhere between 1 and 3. So that puts the player count at between 233 and 700.
700 seems ridiculous, but then again, 3 hours/day as an average also sounds ridiculous, so something's wrong somewhere.
